Epidermal growth factor nanoconjugates comprising quaternary ammonium surfactants and cholesterol

Abstract

Introduction: It has been suggested that topical treatments for complex wounds with exogenous epidermal growth factor affect their bioavailability and effectiveness. Consequently, novel products that can overcome the limitations faced by these treatments are desirable.

Objectives: To develop a nanoformulation whose components and supramolecular structure would increase epidermal growth factor bioavailability at the wound site while remaining biocompatible with the wound.

Methods: The development and evaluation of epidermal growth factor nanoconjugates was carried out in four stages: nanoformulation design, synthesis by a novel supercritical fluid method, physicochemical and biophysical characterization, and evaluation by in vitro biological assays.

Results: Non-covalent epidermal growth factor nanoconjugates comprising quaternary ammonium surfactants and cholesterol were synthesized. These compounds are novel in the context of lipid systems and outperform traditional methods for preparing lipid systems. Characterization of the system using a broad battery of state-of-the-art analytical techniques or better demonstrated that the system exhibits appropriate values for the main critical quality attributes: stability, epidermal growth factor entrapment and release, protease protection, regenerative activity, antimicrobial activity, biocompatibility, and localization capability.

Conclusions: A nanotherapy based on epidermal growth factor nanoconjugates composed of cholesterol and Cetyltrimethylammonium bromide was developed, combining two pharmacological activities: they are a promising vehicle for topical delivery of epidermal growth factor improving the protein's bioavailability and efficacy, and exhibiting intrinsic antimicrobial properties due to their membrane composition.
